Studies on the Toxin Production of the Shiga Bacilli

چکیده

1. The S, R, and R(n) variants of the Shiga bacillus are equally toxic. 2. The effect of the toxin upon rabbits is the same, whether it is derived from filtrates of broth cultures (3 to 6 days old), or is obtained by autolysis of the killed bacteria, grown on agar surface. Rabbits show in both cases prostration, loss in weight, paralysis, and diarrhea. 3. When the toxin is heated to 80 degrees C. for 1 hour, its poisonous effect nearly disappears, but its immunizing ability is unaltered. This heated toxin induces a formation of antitoxin, which can protect against the unheated toxins. 4. The anatomical changes observed in the spinal cord (degeneration of the motor neurons) and in the cecum (hyperemia and hemorrhages) are in agreement with the statements of previous authors. Furthermore, the toxin causes hyperemia and hemorrhages in the heart, hyperemia and degeneration in the kidneys and the liver.

Studies on the Bacteriophage of D'herelle

1. The production of exotoxin and of endotoxin by normal Shiga dysentery bacilli and by strains resistant to Laudman phage was found to be the same. 2. The presence of phage did not alter toxin production by the resistant organism.

Detection of Shiga toxin-producing Escherichia coli (STEC) in faeces of healthy calves in Mashhad, Iran

The aim of this study was to identify virulent Shiga toxin-producing Escherichia coli (STEC) strains isolated from faecal samples of 100 clinically healthy calves. In the present study, a total of 100 Escherichia coli (E. coli) isolates from clinically healthy calves belonging to 6 different farms located in Khorasan Razavi province, Iran, were examined for presence of virulence genes character...

Prevalence, Molecular Characterization and Serology of Shiga toxin-Producing Escherichia coli Isolated from Buffaloes in West Azerbaijan, Iran

This present study is the first to report the presence of Shiga toxin-producing Escherichia coli (STEC) in buffaloes in Iran. A total of 360 fecal samples were collected from buffaloes from different regions in the west Azerbaijan province of Iran and cultured for the isolation of E. coli using routine biochemical tests. From the fecal samples, 340 E. coli were isolated and, of these, 26 STEC i...

In vivo Characterization of Fusion Protein Comprising of A1 Subunit of Shiga Toxin and Human GM-CSF: Assessment of Its Immunogenicity and Toxicity

Background: Most cancer cells become resistant to anti-cancer agents. In the last few years, a new approach for targeted therapy of human cancer has been developed using immunotoxins which comprise both the cell targeting and the cell killing moieties. Methods: In the present study, the recombinant Shiga toxin A1 subunit fused to human granulocyte-macrophage colony stimulating factor (A1-GM-CSF...

Expression of a Chimeric Protein Containing the Catalytic Domain of Shiga-Like Toxin and Human Granulocyte Macrophage Colony-Stimulating Factor (hGM-CSF) in Escherichia coli and Its Recognition by Reciprocal Antibodies

Fusion of two genes at DNA level produces a single protein, known as a chimeric protein. Immunotoxins are chimeric proteins composed of specific cell targeting and cell killing moieties. Bacterial or plant toxins are commonly used as the killing moieties of the chimeric immunotoxins. In this investigation, the catalytic domain of Shiga-like toxin (A1) was fused to human granulocyte macrophage ...

Prevalence of Multidrug Resistant Shiga Toxin-Producing Escherichia coli in Cattle Meat and Its Contact Surfaces

Background: Shiga toxin-producing Escherichia coli (STEC) are group of E. coli causing bloody diarrhea. The goal of this survey was to determine the prevalence of multidrug resistant shiga toxin-producing E. coli in cattle meat and its contact surfaces. Methods: Swab samples (n=120) were randomly collected from meat and contact surface of butchery shops in Sharkia province, Egypt. Prevalence o...
